responsibilities:
- Design and construct complex expression vectors using Gibson assembly, Golden Gate cloning, and site-directed mutagenesis.
- Develop and implement automated workflows for high-throughput vector construction on liquid handling platforms.
- Perform molecular characterization of cell lines using ddPCR, Sanger sequencing, and restriction mapping.
- Generate, validate, and bank vector constructs for candidate biotherapeutic molecules in mammalian systems.
- Support NGS library preparation for genomic and transcriptomic analysis of CHO cell lines.
- Optimize and troubleshoot molecular biology protocols to enhance reproducibility and process efficiency.
- Maintain detailed electronic laboratory notebooks and present technical data to global cross-functional teams.
qualifications:
Required Skills
- PhD in Molecular Biology, Biochemistry, Microbiology, Bioengineering, or a related discipline (0-3+ years of experience).
- Expertise in advanced cloning techniques: Gibson assembly and Golden Gate assembly are essential.
- Hands-on experience with DNA construct design, vector engineering, and plasmid production.
- Proficiency in genetic characterization methods including ddPCR, qPCR, and DNA sequencing analysis.
- Ability to work 100% onsite in Framingham, MA.
- This role requires individuals to be fully vaccinated against COVID-19 as part of your job responsibilities. According to the CDC, an individual is considered to be fully vaccinated fourteen (14) days after receiving (a) the second dose of the Moderna or Pfizer vaccine, or (b) the single dose of the J&J vaccine. Fully vaccinated is to be fully vaccinated 14 DAYS PRIOR TO START DATE.
- Experience with laboratory automation and liquid handling platforms (Hamilton, Tecan, etc.).
- Experience with CHO cell culture or mammalian cell line development.
- Familiarity with in-silico cloning software such as SnapGene, Benchling, or Geneious.
- Experience with NGS workflows, specifically Oxford Nanopore Technologies.
- Basic programming/scripting skills (Python, R, or Bash) for data analysis.
